چکیده
Killer lectin-like receptor CD94 on natural killer cells can bind to HLA-E, 2,3-NeuAc-containing glycoproteins, and heparin. Using the glutathione-S-transferase-fused CD94 (rGST-CD94), we investigated its binding to plates coated with 2,3-NeuAc-containing transferrin secreted by HepG2 cells (HepTF) and sulfate-containing glycanconjugated BSA. rGST-CD94 bound directly to several glycans with similar high affinities: rGST-CD94 bound to heparin-BSA, heparan sulfate-BSA, fucoidan-BSA, -carrageenan-BSA, and HepTF with Kd values of 36 nM, 141 nM, 36 nM, 92 nM and 84 nM, respectively. Different mutants of rGST-CD94 were generated by site-directed mutagenesis. Mutants N160A and C166G had reduced binding to sulfated glycans and HepTF, also F114A, L162A, D163A, and E164A showed reduced binding to sulfated glycans with some variability. These results indicated that CD94 interacts with sulfated glycans and 2,3-NeuAc-containing glycoproteins suggesting that glycan binding sites in CD94 partially overlap with the HLA-E binding sites to modulate natural killer cell-dependent cytotoxicity.

Binding affinities of NKG2D and CD94 to sialyl Lewis X-expressing N-glycans and heparin.
Lectin-like receptors natural killer group 2D (NKG2D) and CD94 on natural killer (NK) cells bind to α2,3-NeuAc-containing N-glycans and heparin/heparan sulfate (HS). Using recombinant glutathione S-transferase-fused extracellular lectin-like domains of NKG2D (rGST-NKG2Dlec) and CD94 (rGST-CD94lec), we evaluated their binding affinities (K(d)) to high sialyl Lewis X (sLeX)-expressing transferrin...
متن کاملCharacterization of CD33 as a new member of the sialoadhesin family of cellular interaction molecules.
CD33 is a member of the Ig superfamily that is restricted to cells of the myelomonocytic lineage but whose functions and binding properties are unknown. It shares sequence similarity with sialoadhesin, CD22, and the myelin-associated glycoprotein, which constitute the Sialoadhesin family of sialic acid-dependent cell adhesion molecules. In the present study, we show that CD33 is a fourth member...
متن کاملPOMGNT1 Is Glycosylated by Mucin-Type O-Glycans.
Protein O-linked mannose β1,2-N-acetylglucosaminyltransferase 1 (POMGNT1) is a Golgi glycosyltransferase that catalyzes the formation of the N-acetylglucosamine (GlcNAc) β1→2Man linkage of O-mannosyl glycan. POMGNT1 is not modified by N-glycans because there are no potential N-glycosylation sites; however, it is not clear whether POMGNT1 is modified by O-glycans. To determine whether POMGNT1 is...
متن کاملN-glycans modulate the function of human corticosteroid-binding globulin.
Human corticosteroid-binding globulin (CBG), a heavily glycosylated protein containing six N-linked glycosylation sites, transports cortisol and other corticosteroids in blood circulation. Here, we investigate the biological importance of the N-glycans of CBG derived from human serum by performing a structural and functional characterization of CBG N-glycosylation. Liquid chromatography-tandem ...
متن کاملAnalysis of Recombinant CD24 Glycans by MALDI-TOF-MS Reveals Prevalence of Sialyl-T Antigen.
CD24 is a glycosyl-phosphatidyl-inositol linked glycoprotein expressed in a broad range of cell types and is heavily glycosylated. It has been found to be over expressed in cancers and tumors and is also a costimulatory molecule. Therefore, this study was carried out to define the structures of the carbohydrates associated with the CD24 recombinant protein. The CD24 glycoprotein's oligosacchari...
